The .AB1 file extension is used for DNA electropherograms. This type of file is created with the Sequencing Analysis Software Bio-Rad, and contains DNA "reads" or sequences from automated sequencers that are used by labs specializing in genetic bio-informatics. The data stored in the .AB1 file includes dye concentration, peak information, sample data, and the actual sequence data. The file is text based and can be opened using a text editor like WordPad.

Structure & Content of AB1 Files

A .AB1 files contains several thousand bytes of data. It starts with an 8 byte preamble that is followed by an arbitrary number of records. Each record consists of four parts: Read Descriptor, Trace Data, Peak Data and Sequencing Data. The Read Descriptor is a 4-byte field, followed by the trace data which consists of multiple 256-byte records. Peak data follows and ends with the sequencing data which is usually 3000-4000 bytes long. In addition to the actual sequence data, .AB1 files also contain additional information, such as quality scores. Other related file extensions are .SCF, BA2, .MSF, and .STK.

Uses of AB1 Files

.AB1 files are mainly used for storing automated DNA sequencing results. These files are typically generated by sequencers that use fluorescence based technologies. They are sometimes referred to as “abif” files, although strictly speaking they are not the same format. Lab technicians also use these files to analyze gene allele characteristics, centromere mapping, base pair analysis, etc.
